铁皮石斛根、茎和叶中内生真菌分离与鉴定(1)

     摘要:目的 分離鉴定铁皮石斛的内生真菌,为铁皮石斛的开发利用提供依据。方法 以多种培养基为基础分离培养基,分别从铁皮石斛的根、茎和叶中分离内生真菌,采用平板划线法对分离得到的菌株进行纯化,并观察其菌落形态。利用形态学、生理生化分子特性及分子生物学手段鉴定内生真菌。结果 分离得到7株铁皮石斛内生真菌,根、茎和叶中的真菌种类和数量各不相同,仅X1菌株同时存在于根、茎和叶部位。通过对性状稳定的3株真菌进行分子生物学鉴定,XJ1与链格孢菌Alternaria sp.的16S rDNA序列有99%的同源性,XY6与链格孢菌Alternaria sp.的16S rDNA序列有93%的同源性,X1菌株与好食链孢霉菌Neurospora africana的16S rDNA序列有99%的同源性。结论 本研究分离并鉴定了铁皮石斛根、茎和叶中的内生真菌,可为铁皮石斛资源的开发利用提供依据。

Isolation and Identification of Endophytic Fungi in Roots, Stems and Leaves of Dendrobium officinale

    Abstract: Objective To isolate and identify the endophytic fungi in different tissues of Dendrobium officinale; To provide basis for development and utilization of Dendrobium officinale. Methods The endophytic fungi was isolated from the roots, stems and leaves of Dendrobium officinale by using various basal culture media. The isolated strains were purified by the plate streaking, and their colony morphology was observed. Endophytic fungi were identified by morphological, physiological and biochemical molecular characteristics and molecular biological methods. Results Seven strains of endophytic fungi were isolated. The quantity and species of microorganisms differed in roots, stems and leaves. Only X1 existed in roots, stems and leaves at the same time. Three strains of fungi with stable characters were identified by molecular biology. By molecular bioassay of three fungi with stable traits, XJ1 had 99% homology with the 16S rDNA sequence of Alternaria sp.; XY6 had 93% homology with the 16S rDNA sequence of Alternaria sp.; X1 strain had 99% homology with the 16S rDNA sequence of Neurospora africana.
